â€Å"The Metamorphosis† unfolds into a novella where the four main characters develop over the course of the story. Their development is accredited to once single event which happens to the main character, Gregor Samsa. Gregor becoming a gigantic bug not only changes his perspective of life, literally, but also impacts his family dynamic in a negative way which will never be restored. With this result of the family shifting, Gregor feel he is the cause for all the pain among family. Gregor was the bread winner for the family, practically the man of the house, yet his father is still alive and well. When money was tight and stress was on the rise, he took on the role of becoming the main source of income for the family. Even though it meant talking on a job he is not overly thrilled about, he did so out of the kindness of his heart. Generosity is in his character and he loved helping out where he could. This made him feel like he was a valuable member of the family; one who c ould never be replaced or forgotten. Eventually earning the income became a burden he felt he had to do; he needed to be able to support his family (Charter p.479). However, Gregor remains calm and accepting despite all his hardships. Not once does he complain or question the transformation. Personality wise, Gregor stays true to his core character but he does struggle with how he now identifies himself. A constant theme of infatuation progressively is displayed throughout Romeo and Juliet, starting with an innocent desire for true love, and ending in death. Romeo and Juliet are the main characters that display the obsession through dialogue. The first sign of infatuation began with Romeo. He claimed, â€Å"love, whose view is muffled still, should without eyes, see pathways to his will† (Shakespeare 20). Romeo is obsessed with the idea of love, and is convinced that love controls you. He ends up getting confused and makes statements about how love is â€Å"heavy and light†, â€Å"bright and dark†, â€Å"hot and cold,† (Shakespeare 20). Stuck in the Middle Films Stuck in the Middle Joe Egan and Gerry Rafferty were a duo known as Stealers Wheel when they recorded a Dylanesque pop hit, "Stuck in the Middle With You", in April of 1974. The single reached number five on the charts - little did they know that eighteen years later it would become a cult favorite. In 1992 Quentin Tarantino, a little known writer/director, took the Cannes film festival and the world by surprise with his motion picture Reservoir Dogs. The movie is about the difficulties that occur when five "master" criminals are hired by a crime king pin named Joe to pull off the biggest diamond heist of the century. Stuck right in the middle of the movie, the Egan/Rafferty hit is played as a introduction to one of the best or worst torture seens ever in the history of movies. It depends on how you look at it. I'll set-up the scene in the movie where it is being played, try and follow me... The five criminals hired go by color-coded names . During the heist the cops show and things got out of control. Two of the robbers were shot and killed after Mr. Blonde, the "on the edge" gangster started shooting up the place when an employee triggered the alarm. Mr. White and Mr. Orange (an undercover cop) escaped the scene and headed for the hideout where all the men were supposed to meet. On the way to the hideout Mr. Orange was shot, he was bleeding severely but the injury was not life threatening. Shortly after their arrival, Mr. Pink met with them and they all anxiousley waited for Mr. Blonde. Mr. Blonde, acting cool and unaffected by the mornings events, made his entrance. After being questioned by Mr. White about why he went psycho in the store, Mr. Blonde called them out to see a "surprise" he had in his trunk. Mr. Blonde in an effort to find out how the police heard about the robbery in advance had kidnapped a police officer. They carried the man into the warehouse and after tying him to a chair Mr. White and Mr. Pink commenced beating the hell out of him. They Asked him to tell how the police knew of the heist, he said he knew nothing and after beating on him some more, Nice Guy Eddie came in. He was Joe's son and told Mr. White and Mr. Pink that they would have to come with him to ditch the cars. Mr. Blonde was told to stay and keep an eye on the cop and the injured Mr. Orange. My idea is that the following scene was written by Director Tarantino choreographed to the song by Stealers Wheel. Rather than the norm where a scene is written and the music is picked thereafter. As I describe the scene I will give the lyrics to the song and show how they correspond to the characters actions in the scene. Mr. Blonde starts talking to the cop, who still insists he knows nothing. The lyrics to the song begin; keep in mind that I am suggesting that the words are what the cop is thinking. Well I don't know why I came here tonight. I got the feeling that something ain't right. I'm so scared in case I fall off my chair, and I'm wondering how I'll get down the stairs. Clowns to the left of me, Jokers to the right... Here I am stuck in the middle with you. The words being sung tell us that the cop is wondering how he got into this situation, he knows something is up, and he is scared that he may not be able to make it through the rest of this torturous interrogation alive. The clowns are his fellow officers who allowed the situation to get so out of control, and the jokers are Mr. White and Mr. Pink. And now he is stuck in the middle of this whole ordeal with the most cynical, evil, and hardest criminal of the bunch....Mr. Blonde. Class, society, and politics in the home, on an interpersonal level among the characters, are themes of outmost importance in the novel â€Å"Sense and Sensibility† by Jane Austen. In this short essay, we shall discuss these themes in relation to the characters, as presented in the novel. Austen presents these social conditions throughout the story, as they were dominant in the society of her time. Laws surrounding inheritance, and property, social etiquette, and money matters in a time were gender limitations were very apparent, signified a person's position in the social scale. The Dashwood women, as they appear in â€Å"Sense and sensibility†, suddenly find themselves in a humiliating situation, when the mother, Mrs. Dashwood, becomes a widow. They were immediately cast into a dire situation. They were deprived of their estate and income when Mr. John Dashwood, became the legitimate heir of all of Mr. Dashwood's fortune, and decided not to support the Dashwood women financially. Mrs. Dashwood and her three daughters, Marianne, Elinor and Margaret had no entitlement to any of Mr. Dashwood's wealth. Having no income resources, they depended on John's charity for support. This was a standard practice of the eighteenth century legal system. In Victorian times, women had, by law, no rights on property. They were seen as dependent on men not only to survive, but also in order to keep their status and respectability. Class divisions, social status and the struggle for its gaining and maintenance are important themes in the novel. All characters in the story come from wealthy, upper class backgrounds. They all belong to high society and their interests and occupations imply this. Sir John Middleton seems to symbolise the best of upper class society. Like the majority of upper class people, his occupation and marital status defined his high position within his social circle. His occupation is hunting, his wealth is inherited, and his wife's only occupation is to raise children. Lady Middleton gathers all the characteristics of the ideal upper class woman: she stays in the home, marries, and is very formal and extremely polite, perhaps to the point of irritation. Characters constantly seem to compete for financial and social power. Inherited wealth is presented as the trademark of high social status. The Dashwood women become deprived of both. As a result they are driven into hardship when they have to give up their house in Norland and all its comforts and luxuries and move to smaller, less luxurious premises in Devonshire. By using this relocation theme, Jane Austen could imply their descent from a high social position to a lower one. It seems almost as if the Dashwoods were â€Å"expelled† from Norland's â€Å"paradise† to Barton Cottage's â€Å"hell† merely because of their female nature. In Austen's world, being a woman in a man's world is a harsh, cruel reality. Marriage for status is an issue of great importance in Sense and Sensibility†. It was seen as a common way in order to gain status. What is more, it was not seen as a choice, but a necessity. Men would seek status through inheritance and/or a wealthy marriage. Edward Ferrars is a typical example of this type of a man in the story. Women would not only opt for handsome and educated men, but mainly for wealthy ones, like Lucy Steele. Wealthier women were also more likely to be legitimate for marriage depending on the dowries they could offer. This put Marianne and Elinor in immediate disadvantage to other women in the novel (i.e. the Steele ladies), and subsequent pressure, in terms of their legitimacy. Their strife for successful marriages mainly stemmed from a desire for social settlement and a restoration of their loss of status. However, in Marianne's case, the main motive for marriage seems to be love (sensibility) and not money (sense), unlike Elinor. Nevertheless, she seems to be almost pushed in a â€Å"proper† marriage with Colonel Brandon in the end. Men seem to have a lot more space for manoeuvering through suppressive social rules than women do in the novel. Society appears to allow them more freedom in marriage and age issues; Colonel Brandon makes a good example in this case. His advanced age did not eventually stop him from getting married to Marianne. In Jane Austen's world as presented in the novel, social integration and acceptance depends on one's status and his/her sensible, controlled behaviour. There seems to be very little room for individuality and deviance. Elinor, Edward Ferrars and the Middletons make fine examples of sensible, controlled characters. As Austen describes: â€Å"Elinor †¦possessed a strength of understanding, and coolness of judgment†¦She had an excellent heart; her disposition was affectionate, and her feelings were strong: but she knew how to govern them† (p4-5). Edward's propriety and the Middletons' formality also signify their extreme level of political correctness and adherence to strict codes of conduct in order to be accepted in society. Social etiquette, formal code of conduct, and discretion to the point of concealment of one's feelings, are also important features in the novel. Desire for social advancement is also evident. Mrs. John Dashwood, for example, was fast to install herself â€Å"mistress of Norland†. In the struggle for financial and social power, women appear to act within their own sphere of action: the home. Women like Mrs. J. Dashwood and Mrs. Ferrars, use domestic and financial politics to ensure their control over situations, within and outside their home environment. Financial and domestic politics seem to be the only means women can use to have their own way in the world of the novel. Ironically enough, even though the story is set in a male-dominated society, the male characters possess little power over women like, for example, Mrs. J. Dashwood and Mrs. Smith and Ferrars. In terms of politics in the home environment, it almost seems as if under the surface of a male-dominated society, what is actually going on is beyond what eyes can see. Austen successfully conveys this idea.

Mutagenicity of Makahiya (Mimosa Pudica) Decoction Chapter I BACKGROUND OF THE STUDY Plants have been a valuable source of natural products for maintaining human health, and the use of herbal plants as pharmacological treatments of diseases began long ago. A lot of people who are below the poverty line cannot afford the high cost of commercial synthetic medicines from drugstores and thus opt to use local plants within their reach since these can be obtained readily, cheaply and entail easy preparation and application (Racadio, 2008). Mimosa pudica Linn, which is known as â€Å"Makahiya† in Filipino, is a popular ornamental plant, as its leaves fold up when stimulated by touch, heat or wind (Balag-ey, 2009). It is considered as a common weed that abundantly grows here in the Philippines. Aside from ornamental purposes, local folks use the decoction of the plant for relief from common illnesses, such as asthma, dysentery, dysmenorrhea, rheumatoid arthritis, cough, fever, sore throat and a lot more (Castillo, et. al. 2005). One of the major concerns these days on the safety of plant extracts, is their mutagenicity. Mutagenicity refers to a chemical or physical agent’s capacity to cause mutations or genetic alterations. The identification of plant extracts with chemicals or compounds capable of inducing mutations is crucial since mutagenic compounds can potentially induce cancer (Ming, 2014). Makahiya has been discovered to contain mimosine. Mimosine is a toxic alkaloid that possesses an antimitotic activity that blocks the cell cycle and inhibits DNA synthesis (Xuan, et. al., 2013) thus might cause genomic instability. This may be a factor that leads to the onset of genetic mutations. So far, a study done by Jadhav et.al in 2013 shows that Mimosa pudica seeds, after S9 metabolic activation were found to be mutagenic and significant. Based on this premise, the researchers conducted a similar study, but instead of focusing on just the seeds of the plant, the researchers used the whole plant. Most importantly, this study utilized the decoction extract of the plant since this is the one being used by the local folks. Furthermore, the Muta-chromoplate kit was used instead of the standard Ames test used by Jadhav et.al. Objectives of the Study This study aimed to identify the mutagenicity of Makahiya (Mimosa pudica) decoction and fresh ethanolic extract using Muta-ChromoplateTM kit. Specific Objectives 1. Scope and Limitations The scope of this experimental study was the determination of the mutagenicity of the decoction and fresh ethanolic extract of Makahiya. The herb was gathered in Davao City and the extracts were prepared in Davao Medical School Foundation, Inc. laboratory, Dr. A. Gahol Street, Bajada, Davao City. There was no utilization of animals as testing subjects. Rather, Muta-Chromo Plate Kit with Salmonella tyhimirium TA100 strain, which was based on the reverse-mutation test, known as â€Å"Ames Test† was used and provided by the EBPI Company. Also, the S9 enzyme activation was through the use of a crude liver extract provided in the kit. Theoretical Framework The theoretical framework of this study was based on a test developed by Bruce Ames, commonly known as the Ames test. In the study conducted by Ames et al. in 1975 (as cited in Razak et al, 2007) they used a test that utilizes a bacterial strain of the Salmonella typhimurium as tool to detect mutations. These S. typhimurium strains also have other characteristics that enhance their ability to detect mutations. Mutation in the histidine biosynthesis gene makes the cell wall of the bacteria more permeable to large molecules. Mutation in a gene responsible for proper excision and repair of DNA damage increases their sensitivity to mutagens. The strains of S. typhimurium are known as auxotrophs which mean that they are unable to produce a required nutrient. The test organism cannot synthesize the amino acid histidine, thus, it will not grow unless the nutrient is supplied in the growth media.Auxotrophs are usually produced as a result of a mutation that occurs in a prototroph, a bacterium that is able to synthesize the particular nutrient. The Ames’ test determines the ability of a tested substance to cause a reversal, also called a back-mutation of these auxotrophs to the original prototrophic state. During the test, auxotrophs are grown in glucose-minimal salts agar plates that contained all required nutrients but only trace amounts of histidine and biotin. The auxotrophs are able to grow for several generations until the histidine in the media was exhausted. This time they will stop growing unless they have sustained a back-mutation that has restored their ability to synthesize histidine. Suspected mutagenic substances were tested for their ability to stimulate back- mutations by placement set on the surface of the minimal agar plates previously inoculated with the auxotroph. The test substance diffused into the surrounding media and, if mutagenic, would induce back-mutations which would then allow growth into visible colonies. The more revertants observed near the test substance, relative to experimental controls, the more mutagenic the substance was likely to be. Conceptual Framework The study aimed to determine the mutagenic activity of decoction and fresh ethanolic extract of Makahiya on Salmonella typhimurium TA100 strain which involved substitution, addition, or deletion of one or a few DNA base pairs. The independent variables were the decoction and fresh ethanolic extract of Makahiya. I enjoy cooking because it keeps me happy in my life. Whenever I feel sad or upset, cooking ease that for me in a big way. I started cooking when I was 6, but it was as simple as putting ham and cheese between 2 slices of bread and used my favorite condiment of maynaise and let it heat up for about 30 seconds. When I grew older, my knowledge of food grew to new depths when I learned how to cook American food on the grill when I was 13 and my dad was the sole reason why I got better since he explained to me how to use temperature control. The 1st dish I made on my own without any kind of assistance was Alaskan Cod with Lemon Pepper which I enjoyed very much when it was baked in an oven. Now, in my college years, I evolved into making cursine in many cultures such as tacos from Mexico, crepes from France, peking duck from China, and pizza from Italy using my ingredients I purchased from HEB, Fiesta, or Chinatown in my hometown. Cooking is the only thing in which I know how to do best and I usually cook for my family, relatives, friends, or people whom I do not know. Sometimes, I cook multiple of one thing for homeless people and disturbute them across the city. My least favorite thing to cook is vegetables since I do not like them, but if someone requests them on their plate, I will not be unwilling to cook them. Instead, I would embrace their desires and make it for them. I have invested hours of my time per day to be more knowlegable about different cooking methods from various chefs instead of just one in order to be prepared for different styles of cooking and perferences from guests. My signature dish is pork and leek dumplings with either soy sauce or vinegar or my orange chicken with basil and coconut broth so if you are ever in my area, I will be willing to cook for you.
